I have started my career in Virology since 2005 as a MPhil student and have more than 66 publications in international peer reviewed journals. I was awarded with the Wong Ching Yee Medical Postgraduate Scholarship and the Research Output Prize of the LKS Faculty of Medicine, HKU, during my PhD study. I have been involved in the discovery of an alternative influenza receptor of avian influenza virus in the human upper respiratory tract (Nature Medicine, 2007) and thereby established various mammalian respiratory epithelial explant models for the study of emerging viruses. To align with the idea of One-Health, I have expanded the options of the respiratory organ explant models to multiple mammalian species, including ferret, pig, guinea pig and mouse, to combat with new challenges from the Human-Animal-Ecosystem interface. My contribution to the better understanding of influenza virus tropism in human, the Hong Kong Institution of Science awarded me with the Young Scientist Award 2011. In the same year, I received the Young Investigator Award in the XII International Symposium on Respiratory Viral Infections.
My main publications are on risk assessing the emerging viruses, including influenza virus and coronavirus by studying their tissue tropism and pathogenesis in human and swine respiratory organ ex vivo culture models and well-differentiated primary respiratory cell culture models. As a long-term collaboration with St Jude Children’s Research Hospital and The Pulmonary Centre of the Boston University School of Medicine, I attached as Visiting Scientist in Professor Robert Webster’s Laboratory and in Professor Jay Mizgerd’s Laboratory in the fall of 2011, respectively.
My current research interests in the Department of Paediatrics focus on the interaction of rhinovirus C with the human respiratory epithelia (nasopharynx, bronchus and lung) and immune cells. These will be supplemented with animal models (e.g. mice and pig, collaboration with Prof Joseph P Mizgerd, Pulmonary Center, Boston University School of Medicine), patient-specific organoid model from airways samples (in collaboration with Dr. Jeffery Beekman, University Medical Centre, Utrecht) and human induced pluripotent stem cells derived airway and lung organoids (in collaboration with Prof Darrell Kotton, Center for Regenerative Medicine, Boston University School of Medicine). Ultimately, this assembly of in vitro and ex vivo culture models with high mimicry with the epithelia in human would allow us to address questions on the pathogenesis of human rhinovirus and the associated respiratory disease, particularly the asthma predisposition and asthma exacerbations. I have visited the Department of Paediatrics Pulmonology and the Regenerative Medicine Center Utrecht in July 2018 to foster the collaboration set in March 2017, the CUHK-UMCU Joint Research Laboratory of Respiratory Virus & Immunobiology.
I have obtained a total of 9 competitive grants (3 GRF and 6 HMRFs) with a total funding amount of over HK$8 million. Dr Chan is also an Academic Editor of the journals: Journal of Virological Methods, PLoS ONE, Madridge Journal of Immunology, and the ad-hoc reviewer of Scientific Report, Respiratory Research and Antiviral Research.
Research Interest: The Epidemiology and Pathogenesis of Respiratory Virus Infections, especially in the paediatric population.
