The overall focus of my lab is to define the role of key signal transduction mechanisms that regulate thyroid cancer growth and metastasis, with a specific focus on the Src/Focal Adhesion Kinase (FAK) and MAP kinase signaling pathways. My lab discovered that FAK is overexpressed and phosphorylated in thyroid tumor samples, and that inhibition of the FAK/Src pathway blocks growth, invasion, and metastasis in thyroid cancer preclinical models. We have further shown that FAK/Src signals independently of the MAP kinase pathway, indicating that the FAK/Src pathway may represent a second independent risk factor for thyroid cancer patients, especially in relation to tumor aggressiveness. The goals of my lab are to 1) understand the scaffolding versus kinase functions of FAK and the novel role of nucleolar FAK in thyroid cancer, and 2) to define pathways that promote sensitivity versus resistance to Src inhibition.
For these studies, my lab has developed many relevant in vitro models to study signal transduction methods, as well as relevant in vivo preclinical models to help translate our findings to the clinic. We are one of the few labs in the world that uses an orthotopic model of thyroid cancer to better recapitulate the tumor microenvironment. My lab has also developed the first distant metastasis model for thyroid cancer, to study clinically relevant metastases to the brain and bone. Importantly, I discovered that the majority of thyroid cancer cell lines that had been widely used by labs around the world for the past 20 years were misidentified with cell lines from other tumor types. Through our Challenge grant (1RC1CA147371-01), we have authenticated and characterized a panel of 60 thyroid cancer cell lines, and we are developing new thyroid cancer cell lines and patient-derived xenograft models. My lab is very collaborative, and we are always willing to share reagents, protocols, and model systems. I have also received strong clinically relevant training through the Paul Calabresi Clinical Oncology Scholars Award (K12-CA086913), which has provided me with the necessary skills and knowledge to apply my basic research findings to the clinic.
In 2013, I was recognized with the “Rising Stars” outstanding junior faculty award from the Department of Medicine at the University of Colorado for excellence in research and teaching. I am very committed to training PhD and MD scientists, and have been actively involved in graduate student training since 2012. I have held key leadership roles in the Cancer Biology Graduate Program, including Associate Director, Student Advisor, and Chair of Admissions, and in August of 2021, I became Program Director. In 2014, I received an Outstanding Faculty Sponsor award and in 2019, I received the Dean's Doctoral Mentoring Award, both from the University of Colorado Graduate School.
