RESEARCH PROFILE OF DR. PUERTAS-AVENDAÑO RICARDO ALFONSO
Dr. Ricardo Alfonso Puertas Avendaño, obtained his PhD in Biology in 2016 in the Department of Biochemistry, Microbiology, Cell Biology and Genetics, area of knowledge Cell Biology, by the University of La Laguna (Tenerife, Spain), with the study of the DLK1 protein in the Hypophysis-Adipose Endocrine Axis, in which various techniques were used, including: Transmission Electron Microscopy for postembedding samples, immunofluorescence for confocal microscopy, immunostaining techniques with chromogens, PCR, ELISA, among others. This study demonstrated for the first time the subcellular localisation of the DLK1 protein in adult mouse adeno-pituitary cells and its functional relationship with Leptin (Ob). They also demonstrated in an in vivo model of wild-type and knockout adult mice for DLK1, that adipose cells express all the receptors for adeno-pituitary hormones, and in turn adeno-pituitary cells express the Leptin receptor, as well as Leptin (Ob) itself. Thus, they demonstrated the existence of a new Endocrine Axis, in which the DLK1 protein plays a modulatory role.
After completing his PhD studies in 2016, Dr. Puertas Avendaño joined the research group of the Cellular Neurobiology Laboratory of the Department of Basic Medical Sciences, ULL, as a postdoctoral researcher in the project: SAF-2014-52582-R.
Moreover, he has enjoyed 3 postdoctoral stays, two of them in the area of Electron Microscopy at the Faculty of Medicine of the University of Las Palmas de Gran Canaria.
It should be noted that Dr. Puertas Avendaño obtained a competitive research grant from the Fundación Canaria "Dr. Manuel Morales" to carry out his third postdoctoral stay at the Institute of Neurological Disabilities of the Faculty of Medicine of the University of Castilla-La Mancha. During this postdoctoral stay she actively participated in the European Human Brain Project.
In addition, and within the research profile of Puertas Avendaño, it is worth highlighting his teaching and research experience during five years at the Faculty of Dentistry of the Santa María University in Caracas, Venezuela. There, he taught in areas of knowledge such as Microbiology, Biochemistry, Molecular Biology and Genetics, among others.
Finally, it is important to mention in Dr. Puertas Avendaño's research profile the techniques and large laboratory equipment that he handles with absolute autonomy. As well as the interpretation of results derived from scientific experimentation.
Laboratory techniques.
Flat inlay techniques for pre-embedding with one or two markers.
Flat or block embedding techniques for post-embedding with one, two or three markers.
Histoblot (technique that provides very high information on gene expression in embryonic and non-embryonic development in fresh tissue sections).
Western blot.
Dot blot.
Co-immunoprecipitation.
.- PCR, real time
ELISA
Cell culture techniques (primary cultures and cell lines).
Immunohistochemistry and immunocytochemistry of fluorescence or chromogens (DAB, chloro-naphthol or carbazole red, etc.).
Among other techniques...
Large equipment.
Autonomous operation of the Transmission Electron Microscope, TEM-100.
Ultramicrotome (Dr. Puertas Avendaño is currently qualified and authorised by the Electron Microscopy Service of the SEGAI for the autonomous use of the ultramicrotome for the preparation of biological samples. Doc Nº 82).
