Bio
My research interest is focused on the biophysical principles and the biological concepts that underly communication between proteins. In particular, we focus on interactions mediated by short motifs, peptides, which are transient and easily modulated.
By developing state of the art tools for the structural modeling of these interactions at atomic resolution (e.g. Rosetta FlexPepDock, and PIPER FlexPepDock), we wish to make these interactions accessible to fine-tuned, structure-based manipulation via designed competing peptides and peptiderived peptidomimetics.
We have repeatedly generated top-accuracy models of peptide-protein interactions within the CAPRI community - wide protein docking experiment, and our protocols have been the motivation for the development of many additional approaches to peptide docking.
The substrates identified using structure-based peptide binding predictions (Rosetta FlexPepBind) have allowed us to identify new substrates of post-translational modifier enzymes (writers and erasers), as well as substrates of peptide binding domains (readers). Integration of these new connections in the cellular protein-protein interaction networks can reveal new regulatory modules and new biological function.
Many of the peptide-mediated interactions tend to occur as repeats, making it possible to achieve high binding affinity together with easy regulation. A main focus of interest in our group is to detect strategies that integrate information from different peptide-protein interactions into a complex, tightly regulated response.
Over the years, I have had the pleasure to work on protein-protein interactions with an amazing group of interacting researchers at all stages of their scientific career. I believe that a diverse group of scientists that includes members form a wide range of different background is the optimal way to advance science, and society.
