My research interests are focussed on signal transduction in phagocytes, mainly neutrophilic granulocytes. I work on the microbicidal functions of the neutrophil: phagocytosis, degranulation and production of reactive oxygen species (ROS) by the NADPH oxidase. These functions are regulated by G-proteins and GPCRs, immunoblobulin receptors, intracellular calcium and phosphoinositides. I investigate the dynamic aspects of these events by biophysical techniques such as electrophysiology (patch-clamp) and various types of fluorescence microscopy. In recent years, my group mainly used fluorescent proteins tagged to subunits of the NADPH oxidase to investigate the assembly and activation of the enzyme in live cells. In parallel, we developed methods to detect the phagosomal ROS-production in real-time.
