A main focus of my lab is to investigate the correlation between systemic metabolism and cancer incidence and progression, with the goal of identifying metabolic dependencies that could be targeted therapeutically in cancer patients. I have several years of cancer research experience, which I acquired during my post-doctoral training and as an independent investigator, working on both transplant and genetically engineered mouse models of lung and pancreatic cancer. As a post-doctoral fellow, I pinpointed a key role for PI3K signaling in determining the sensitivity of tumors to dietary restriction (Kalaany and Sabatini, Nature, 2009). As an independent investigator, I followed up on this finding (Curry et al., Cancer Discovery, 2013) and identified metabolic vulnerabilities in KRAS-driven lung cancer (Xu et al., PNAS, 2018) and pancreatic cancer (Zaytouni et al., Nat Commun, 2017). Findings from Xu et al. indicated that combinatorial targeting of insulin/IGF1 signaling along with autophagy or proteasome inhibitors may represent an effective therapeutic strategy in KRAS-mutant non-small cell lung cancer. Findings from Zaytouni et al. indicated a dependency of obesity-associated pancreatic cancer on the urea cycle, particularly the extra-hepatic isoform of arginase (ARG2), for the disposal of excess nitrogen. This discovery opened a therapeutic window for specific targeting of fast-growing pancreatic tumors in lean or obese cancer patients. In a recent manuscript (Tsai et al., PNAS, 2021), we identified non-genetic adaptations to nutrient deprivation in pancreatic cancer and highlighted glutamine synthetase enzyme, as a dependency that could be targeted therapeutically in pancreatic cancer patients. In work still in progress, we optimized genetically engineered mouse models of lung and pancreatic cancer for the study of cachexia or body wasting, and are investigating the metabolic alterations that accompany this multi-organ wasting syndrome, how to prevent it or reverse its fatal course of action in cancer patients.
