Dr. Michele Giannattasio was born in Milan, Italy, the 31st March 1972. After his bachelor studies in biological sciences at the unversity of Milano, he obtained his master degree in molecular biology from the University of Milan in 1998, being trained in the laboratory of Prof. Paolo Plevani, Prof. Giovanna Lucchini and prof. Maria Pia Longhese at the department of genetics and microbiology. He used the budding yeast Saccharomyces cerevisiae as model organism to study the role of the p34 RPA subunit in DNA repair. During his PhD in genetics sciences, which he pursued in the Prof. Marco Muzi-Falconi and Prof. Paolo Plevani laboratory at the department of Biomolecular Sciences and Biothecnology of the University of Milan in 2002, he focused on the molecular mechanisms underlying the activation of the DNA damage checkpoints in non-replicating budding yeast cells. His studies uncovered that DNA damage checkpoint activation after UVC irradiation in non-replicating yeast cells requires DNA lesion incision by nucleotide exicision repair (NER) and ssDNA gap extension by Exo1. During these studies, in 2007 Michele was awarded with an EMBO short-term fellowship as a visiting researcher in the Dott. Philippe Pasero laboratory at the institute of genetique humaine in Montpellier (France), where he used DNA combing in non-replicating yeast cells to visualise the long patch NER.
In the following years, Michele discovered that specific histone post-traslational modifications are required for the DNA damage checkpoint activation after UVC irradiation in non-replicating budding yeast cells. In 2006, he received a permanent position as technician at the University of Milan (department of Molecular Biosciences) in the Prof. Marco Muzi-Falconi and Prof. Paolo Plevani laboratory. Michele’s interest in the use of transmission electron microscopy (TEM) for the study of chromosomal DNA intermediates started in 2008 when he utilised this technique to visualise long ssDNA gaps generated during Nucleotide Excision Repair (NER) in non-replicating budding yeast cells (those EM results were published in 2010). In the same year (2010) Michele became staff scientist at IFOM (the FIRC institute of molecular oncology foundation) where he continued to apply transmission electron microscopy techniques to the visualisation and structural analysis of chromosomal DNA intermediates in collaboration with several IFOM research groups (among them dott. Branzei Dana and Prof. Marco Foiani laboratories). In 2016 Michele moved to the department of Oncology and Hematology-Oncology of the University of Milano. From 2010 at the IFOM institute, Michele contributed to the isolation and structural analysis of DNA damage tolerance and DNA replication intermediates and was awarded with a second EMBO short term fellowship (2012) to visit the electron microscopy research centre of the university of Zurich-Irchel Park (deputy director dott. Andres Käch) and the laboratory of Prof. Massimo Lopes at the IMCR institute-Irchel park-Zurich. During this research experience in Zurich Michele learned and applied several TEM techniques and used several electron microscopes and sample preparation machines. Michele was then able to import those TEM techniques for the analysis of single molecules (nucleic acids and proteins) to IFOM and make them available to use to IFOM scientists. In 2016 Michele became head of the IFOM electron microscopy technological development unit (TDU) (DNA/Single Molecules Visualisation Unit).
