After completion of her PHD thesis at Paris Saclay University, Dr. Maryam Mehrpour joined French national Institute of health and medical research (INSERM ). Her work was about cloning, sequencing and the role of inter-alpha-trypsin inhibitor family proteins in lung cancer. She introduced a new model of spontaneous lung metastases detection in animal. This model was patented. Maryam Mehrpour group demonstrated that inhibition of tumor growth and metastatic spreading by overexpression of inter-alpha-trypsin inhibitor family chain.
Then, she spent as guest scientist one year at the Scripps Research Institute (TSRI), La Jolla, CA, USA to study regulation of apoptosis induced by death domain receptor in leukemia cells.
She then joigned Gustave Roussy Institute where she started research on molecular mechanisms of tumor resistance to cell death in human cancer cells. She demonstrated the critical role of prion protein and cFlip in tumor resistance to cell death (Hamai, 2005). At IGR Dr. Mehrpour pioneered the role of prion protein in cancer biology (Diarra-mehrpour, 2004; Meslin 2007). Then as invited professor to Chinese academy of sciences in Beijing, she studied cancer stem cells (Du lei 2008, 2010). She then studied the molecular mechanism of apoptosis and autophagy in cancer cells at Institut Necker Enfants Malades in Patrice Codogno laboratory. She is focusing about the role of autophagy in self-renewal of cancer stem cells (CSCs) and how to eradicate cancer stem cell in breast, prostate and brain cancer. Maryam Mehrpour group demonstrated that autophagy is critical for self-renewal and tumorigenicity of Breast CSCs (Gong, Beauvy 2013) . They then demonstrated that Anti-CSC activity of Salinomycin is associated to autophagic flux inhibition (Wen, Hamaï 2013). Recently, as part of a collaborative work they provide evidence that a synthetic derivative of salinomycin, which we named ironomycin (AM5), exhibits a more potent and selective activity against breast CSCs in vitro and in vivo, by accumulating and sequestering iron in lysosomes, and by activating of cell death pathway consistent with ferroptosis. These findings reveal the prevalence of iron homeostasis in breast CSCs, pointing towards iron-mediated processes as potential targets against these cells (Mai, Hamaï 2017). She published more than 80 paper in PNAS, Cancer Res, Oncogene, autophagy, Nature Rev Mol Cell Biol and Nature Chem…. Since January 2021, She is retired but continued to follow some academic activities.
