I am currently a postdoc fellow in the Tumor Biology Department at the Moffitt Cancer Center. During my PhD in pharmaceutical chemistry, I synthesized multiple series of small molecules as inhibitors of different enzymes involved in the tumor microenvironment, such as matrix metalloproteinases and carbonic anhydrase isoforms. Preliminary docking calculation studies predicted the successful chemical structure features of optimized enzyme inhibitors, whose synthesis was realized in Dr. Paolo Tortorella pharmaceutical –chemistry lab in University of Bari, Italy. Compounds with high purity were obtained in grams quantity thanks to optimization of the synthetic and analytic techniques such as HPLC, MS-SPECT and NMR. In 2013 I started a post-doc in Dr. Lynch lab (Tampa, FL) currently focused on elucidating the role of matrix metalloproteinases (MMPs) in the metastatic bone microenvironment. The combination of my chemistry and biology skills is actually directed in investigating the role of prostate and breast cancer cells in the bone. The presence of cancer cells in bone induces a vicious cycle between tumor cell growth and bone resorption. The contribution of Dr. Lynch in several publications identified that particular MMPs have a pivotal role in regulating the bioavailability and activity of key factors such as TGF-β and PTHrP, promoting the recruitment and activation of osteoclasts, with consequent destruction of hydroxyapatite matrix and release of bone matrix sequestered growth factors that stimulate the proliferation of cancer cells thereby completing the vicious cycle. We realized in vitro and in vivo evaluations thanks to animal models of prostate and breast cancers to bone metastasis. The Lynch lab has extensive expertise in the imaging of bone lesions (microCT, microSPECT, Faxitron-Xray) and in validating the obtained images with histology and histomorphometry. The identification of the contribution of some MMPs in the vicious cycle and the availability of selective MMP inhibitor molecules capable to target the bone are allowing me to translate the use of those molecules in the clinic.
