Anton is a leading expert in 1) stable isotope methodology measuring glucose, fatty acid and protein kinetics in living human beings and 2) confocal immunohistochemical microscopy methods to stain in sections of skeletal muscle the key enzymes that play a role in insulin resistance mechanisms and visualise their spatial distribution and co-localisation with mitochondria, lipid droplets and the microvascular endothelium. Anton's research has generated compelling evidence that insulin resistance occurs both in muscle fibers and their microvasculature. In obesity, metabolic syndrome and ageing insulin mediated dilation of the muscle microvasculature is impaired due to failure of insulin to activate eNOS (ser1177-P), quenching of NO by superoxide anions (NADPH oxidase) and excessive endothelin-1 production. As a result of impaired arteriolar dilation muscle fibres are starved of nutrients in well-fed bodies and meal derived glucose, lipids and amino acids are deposited in non-muscle tissues such as liver and adipose tissue. Recent research has also shown that high intensity intermittent (HIIT) training and endurance training are equally effective in improving insulin resistance of both muscle fibres and their microvasculature with the effects of HIIT being achieved in a fraction of the time. Anton is Editorial Board member of AJP Endo&Metabolism, Adviser of ECSS Executive Board and Honorary Fellow of The Physiological Society (UK).
