Research Line
Our research is focused in the heterotrimeric G protein signaling pathways involved in the neural crest (NC) induction and migration in Xenopus embryos. The neural crest (NC) is a transitory cell population that is induced at the border of the neural plate between the neural and non-neural ectoderm. It gives rise to a range of cell types via differentiation, including craniofacial cartilage and bone, pigment cells and neurons of the peripheral neural system, among others. Later during development, these cells undergo an epithelial to mesenchymal transition (EMT), which is characterized by the acquisition of motility. Specifically we are interested in the study of the molecular mechanisms by which heterotrimeric G proteins regulate the remodeling of actin microfilaments (AM) and microtubules (MT), responsible for the cell shape, adhesion and motility responding to chemical and mechanical cues. Crosstalk between AM and MT relies on small GTPases from the Rho family and other components from the both cytoskeletal networks, which control cell polarity, actin polymerization, and actomyosin contractility. MT are also involved in trafficking, delivering molecules to the cell leading edge, all of them involved in mechanosensing regulating focal adhesions (FA) dynamics, velocity, persistence and directionality of migration. In order to address this studies we use in vitro fertilization, microinjection to perform loss and gain of function analyses and transgenesis, explants for in vitro cultures and transplants, and to analyze cell behavior we use traction force microscopy and high resolution imaging.
