For the past seven years, I have worked in the field of respiratory cell biology with a particular interest in the role of non-coding RNAs in lung diseases such as asthma and COPD. My work has focussed upon how these non-coding RNAs are altered in disease and whether these molecules could be used as novel therapeutic targets. I have published my work in high impact medical journals and have presented both nationally and internationally as well as acting as a reviewer for respiratory medical journals.
Furthermore, I have extensive experience in the supervision of a number of PhD, BSc, MRes and MSc students in the laboratory and during thesis writing at Imperial College London.
My Imperial College Junior Research Fellowship is entitled;
Long intergenic non-coding RNAs in the regulation of airway smooth muscle proliferation and inflammatory gene expression. This includes collaborations with Professor Mark Lindsay (Department of Pharmacy and Pharmacology, University of Bath) and Professor Eric Lam (Department of Molecular Oncology, Imperial College).

My work included research into inflammatory mechanisms in the airway diseases asthma and COPD, with special interest in the biology of airway smooth muscle (ASM). ASM and its interactions in inflammation are at the core of airflow obstruction and bronchial hyperresponsiveness. Regulation of cytokine expression, cellular proliferation and intracellular signaling components are the focus of this area of research, and abnormalities of the muscle in disease are being characterised. The characteristics of the inflammatory response and airway remodelling changes are different in severe asthma and in COPD and are less responsive to corticosteroids; the underlying mechanisms of which were the subject of investigation.
I’ve investigated the role of the gas hydrogen sulfide in ASM proliferation and cytokine release in different asthmatic groups as well as further examining the role of TGF-β and thrombin upon ASM.
The techniques employed for this research included primary cell culture, qRT-PCR, determination of protein levels (ELISA and western) and the manipulation of endogenous hydrogen sulfide levels in vitro.

PhD entitled: “The role of microRNA-146a in the IL-1β-Induced inflammatory response in human lung alveolar epithelial cells”.

Specifically, I was researching the role of miRNAs in the innate immune response in the lung. This has demonstrated that changes in the expression of miRNAs can also regulate acute inflammatory responses in human lung alveolar epithelial cells. Thus, stimulation with IL-1β results in a rapid time and concentration-dependent increase in miRNA-146a and, to a lesser extent, miRNA-146b expression, although these increases were only observed at high IL-1β concentrations. Examination of miRNA function by overexpression and inhibition (siRNA/LNA) showed that increased miRNA-146a expression negatively regulated the release of the proinflammatory chemokines; IL-8 and RANTES. Subsequent examination of the mechanism demonstrated that the action of miRNA-146a was mediated at the translational level and not through the down-regulation of proteins involved in the IL-1β signalling pathway or chemokine transcription or secretion.
Overall, these studies demonstrated that rapid increases in miRNA-146a expression provide a novel mechanism for the negative regulation of severe inflammation during the innate immune response.
