Lisete M. Silva is a Researcher at University of Aveiro (Department of Chemistry), Portugal, since April 2019. She is also an Honorary Research Associate in the Glycosciences Laboratory at Imperial College London. Lisete obtained her BSc degree in Chemistry in 2007 at the Faculty of Sciences, University of Porto (Portugal), and completed her MSc in Chemistry with food specialisation in 2008, at the same University. Her PhD degree in Biochemistry was obtained in 2014 at the University of Aveiro (Portugal), in collaboration with the GlycoLab – Functional Glycobiology at Faculty of Science and Technology, NOVA University of Lisbon (FCT-NOVA) and the Glycosciences Laboratory at Imperial College London.
PhD IN BIOCHEMISTRY: Lisete focused on the structural analyses of lipopolysaccharides (LPS) from clinical isolates of H. pylori by GC-MS and ESI-MSn, and on the development of a bacterial LPS microarray for interaction studies with glycan-binding proteins and human sera.
POST-DOCTORAL STUDIES IN GLYCOBIOLOGY AND BIOCHEMISTRY: Her knowledge on the structural (bio)chemistry of complex carbohydrates, combined with her passion for the glycobiology field and expertise on glycan microarrays, have led her to a Research Associate position in the pioneer glycan microarray team (Glycosciences Laboratory) at Imperial College London in March 2015; and in November 2015 she was named Deputy Microarray Project Leader with responsibility for running, maintaining and troubleshooting the group’s microarray equipment and performing glycan array analyses of proteins, received from national and international collaborators. In parallel, Lisete carried out investigations of different liposomal formulations for lipid-linked glycan probes in neoglycolipid (NGL)-based microarrays with respect to the binding signals they elicit in microarray analyses. These studies led to the construction of a new version of a broad-spectrum microarray of ~ 900 natural or synthetic lipid-linked sequence-defined glycan probes (one of the two largest glycan libraries in the world) for the Wellcome-Trust supported Carbohydrate Microarray Facility, suitable for screening analyses of a variety of carbohydrate recognition systems of biological and medical impact and able to serve the scientific community as international resources. During 2015-2019, Lisete actively contributed and provided leadership support to biotechnology and biomedicine R&D projects (translational research); collaborated with 50+ national and international academic and industrial teams that have resulted in > £30k of revenue for facility services. As an Honorary Research Associate in the Glycosciences Laboratory, Lisete is currently providing consultancy advice to the group regarding microarray equipment and training to the new staff members, assisting with microarray experiments, data analysis and preparation of manuscripts. Back to Portugal in 2019, Lisete has been focused on the development of galactomannan-based microparticles, obtained from different natural sources, for pulmonary insulin delivery. The structural variability of the different galactomannans is being evaluated using conventional analytical tools and complemented with interaction studies with glycan-binding proteins of various recognition systems using glycan microarrays.
SCIENTIFIC ACTIVITY OUTPUTS: Lisete holds 17 publications in relevant peer-reviewed journals of pathogens, glycobiology, molecular cell proteomics and food chemistry and technology and 3 book chapters; 12 oral communications (9 by invitation) presented at international/national conferences. In addition to the Microarray Facility management role over (2015-2019), she also participated in events for dissemination of science through lectures at workshops and performed didactic activities with the public (age 3+).
TEACHING AND SUPERVISION EXPERIENCE: At Imperial College London, Lisete was invited to give Glycobiology lectures to the Immunology MSc students. In addition, she supported PhD students research projects and provided training to professional researchers from various fields in glycan microarrays. In Portugal, to date, Lisete supervised 4 BSc and 1 MSc students in Biochemistry and has given practical classes of the curricular unit of General Chemistry to BSc students.
RESEARCH INTERESTS: Lisete is interested in translational research in the fields of Biochemistry, Biomedicine, Glycobiology, Food and Nutrition and Biotechnology. The main research topics include the following: 1) glycan structural features and bioactivity relationship; 2) designer and construction of microarrays of natural and/or synthetic glycans; 3) to use glycan microarrays for new ligands discovery and to explore glycan-protein interactions in various biological systems, such as pathogen- and microbiota-host interactions that trigger diseases or metabolic processes; 4) development of natural glycan-based products and glycan-based microparticles for drug delivery.
