During my PhD I investigated astrocytic glutamate metabolism and the effects of glutamate dehydrogenase (GDH) expression on energy metabolism and TCA cycle function. It involved intricate work with metabolic studies using 13C and 14C labeled substrates and labeling pattern analysis to identify the involvement of different metabolic pathways and enzymes.
These studies demonstrated an important role of GDH and glutamate and metabolism thereof in maintenance of TCA cycle function and energy production in astrocytes. It was also found that the GDH was an important factor in anaplerosis of TCA cycle intermediates and the utilization of alternative energy substrates for oxidative metabolism during conditions of decreased glucose availability.
Following my PhD I have continued my research within the field of neurometabolism focusing on GDH function and general brain energy metabolism during pathological conditions such as e.g. hepatic encephalopathy and diabetes.
Through recent scientific findings of the group of associate Professor and Chief Physician Miriam Kolko my attention has been drawn towards the importance of GDH in inner retinal conditions. The close interplay of bipolar nerve cells, retinal ganglion cells, and Müller-Cells bears close resemblances with the organization of neurons, synapses and astrocytes in the brain. My previous focus on GDH function and astrocytic energy metabolism is thus a great plateau from which to expand the research on the importance of GDH to pathological conditions of the inner retina.
