My initial research interests and training focused on understanding the pathophysiological mechanisms of traumatic brain injury (TBI). During that training, I learned that TBI is the strongest environmental risk factor for the later development of Alzheimer’s disease (AD). Therefore, for postdoctoral training I sought to expand my study to include investigation of potential mechanistic links between TBI and neurodegeneration and joined Prof. Douglas H. Smith’s group. Since joining his laboratory, I have examined the underlying mechanisms of axonal injury in TBI and how that selective pathology could drive the genesis of protein misfolding similar to pathologies found in AD and AD-related dementias (AD/ADRD). The laboratory has a unique translational approach by examining mechanisms in clinically relevant in vitro and in vivo models and then determining the relevance of data through direct comparisons with post-mortem human TBI neuropathological analyses. Specifically, my work has harnessed the lab’s unique in vitro dynamic stretch injury of micropatterned axon tracts and parallel studies with a clinically relevant in vivo swine model of TBI/ concussion. I also extensively participated in Prof. Smith’s NINDS/NIA Center Without Walls project, the CONNECT-TBI, including leading several projects to characterize the evolving neuropathological changes after TBI in humans. These efforts led to the identification of axonal sodium channel loss and disruption of the node of Ranvier as important pathological substrates of diffuse axonal injury. In addition, I have tested a new hypothesis for pathways leading to axon repair or degeneration, which involves the interaction between axonal microtubule stabilizer, MAP6, and phosphorylated tau (p-tau). My long-term goal is to become an independent academic scientist and lead the discovery of initiating mechanisms of TBI that lead to AD/ADRD.
