In the middle of 1980ies I realized that the molecular understanding of mucus and mucins was very poorly developed and thus I committed myself to change this. By my expertize in mass spectrometry, I first developed methods for mucin glycan analyses, I was part of the cloning and identification of mucin genes. With a commitment to intestinal problems, I first studied the biosynthesis of the MUC2 mucin and its assembly. Studies of the MUC2 mucin lead to the discovery 2008 of two mucus layers in colon and that the inner is normally impenetrable to bacteria. This has opened a new research area that will now be developed further. The last budget period has been very successful with papers showing the packing of mucins, the importance of bicarbonate, and the discovery that the inner mucus layer is penetrable to bacteria in colitis models and ulcerative colitis
