I am currently a principal investigator for Kannalife Sciences. I conduct a platform of assays on neuroprotection and inflammation using primary neural cultures. During my 35-year career, I have been a Section Chief in the Intramural Program at NICHD and a Senior Research Fellow/ Team Leader at Johnson & Johnson Pharmaceutical Research and Development. I have the demonstrated skills required to investigate central nervous system (CNS) pharmacology as evidenced by more than 170 peer-reviewed scientific publications and 26 patents and patent applications. My publication record is focused on evaluating neurotoxic and neuroprotective substances in a variety of CNS and PNS culture preparations. My training in the use of CNS and PNS cultures to study developmental neurobiology was obtained in the laboratory of Dr. Phillip G. Nelson, a pioneer in the use of primary neural culture systems. After I obtained my own Section at NICHD, my laboratory used CNS and PNS cultures to study the neurotrophic properties of neuropeptides and growth factors. Further, as a result of work completed at the NIH, I am the co-inventor of davunetide, a neuroprotective peptide that proceeded to clinical trials for the treatment of progressive supranuclear palsy. In other relevant experience to the current proposal, I was the co-leader of the scientific team at Johnson and Johnson that advanced two novel anticonvulsants through discovery and preclinical testing. Both of these compounds qualified for entry into clinical evaluations. My current work with Kannalife Sciences builds on my background in two specific pharmacological areas as they relate to the action of cannabidiol: neuroprotection and anti-inflammatory activity. Indeed, the identification of KLS-13019 as a potential lead compound has been reported in our recent publication that indicated that the new compound has superior neuroprotective properties to cannabidiol, the cannabis-derived compound with recognized neuroprotective and antiseizure properties. Relevant to the use of siRNA to study drug action in primary hippocampal cultures, my previous work with the NCX-1 target demonstrated the use of high content imaging to localize the target changes in neurites as key to understanding this target in the protective action of KLS-13019.
