Work Summary: I have synthesised diversity of phosphonylpyrazoles, spiropyrazolines and sulfonylpyrazoles via 1,3-dipolar cycloaddition reaction of Bestmann-Ohira reagent.
I have performed total synthesis of Withasomnines and their non-natural analogs from aldehydes and 4-nitro-1-butanol in three steps and the key 1,3-dipolar cycloaddition step is carried out with commercially available TMSCHN2. Withasomnine is a naturally occurring alkaloid and isolated from the roots of Indian medicinal plant Withania somnifera,belonging to the Solanaceae family. It has been used in the traditional Ayurvedic medicine for the treatment of enlarged spleen, migraine and many infections, and as an aphrodisiac. The analgesic as well as CNS and circulatory system depressant properties of Withasomnine and its analogs motivated us to pursue their synthesis.
I have worked on Indo-French project on carbon nanotube(CNT)-metal hybrid catalyzed organic transformations. In this project, I have mainly focused on synthesis of carbon nanotube supported gold nanohybrids for organic transformations. CNTs have recently emerged as a promising alternative to conventional catalyst supports as they provide some unique advantages such as low cost, chemical/thermal/mechanical stability in liquid media, inertness, high specific surface area, chemically tunable topography, and facile reclaim of the metals. CNTs are made of graphene layers rolled-up into a cylindrical form. They can be either composed of a single rolled-up plane of graphene (single-walled carbon nanotubes, SWCNTs) or of multiple concentric layers (multi-walled carbon nanotubes, MWCNTs). Their diameter is in the nanometer scale, while their length can reach several microns. CNTs also enhance the electron transfer rate of many redox reactions and are able to stabilize transient higher oxidation states of supported metals. These intrinsic properties make CNTs extremely attractive as support for small metallic catalyst particles with excellent dispersion and high surface area.
I have synthesised diversity of functionalized imidazoles from Morita-Baylis-Hillman adducts of nitroalkenes and amidines. The imidazoles synthesized by the above methodology have been screened for their activity against the parasite Trypanosoma cruzi that causes Chagas disease. While two of the compounds exhibited activity comparable to that of the standard (benznidazole), one of the compounds was two times more active.

Title: 2-Pyranones-Derived Arenes and Heteroarenes as Biodynamic Agents

Synopsis: The thesis primarily focuses on the interface of synthetic organic and medicinal chemistry. It deals with the design and synthesis of novel privileged class of heterocyclic molecules, biaryls, teraryls (1,2-teraryls, 1,3-teraryls and 1,4-teraryls), phenols, acetophenones, phenylacetones, biaryl ethers, thioethers and organomettalic compounds using 2H-pyran-2-ones as precursors without use of any catalyst and organometallic reagents. It also emphasized the synthesis of polycyclic aromatic and heteroaromatics using 5,6-dihydro-2-oxo-4-sec.amino-2H-benzo[h]chromene-3-carbonitriles as a new type of precursor, elucidation of their structural aspects utilizing various one- and two dimensional NMR spectroscopic methods as well as X-ray crystallography. Additionally the synthesized compound were successfully evaluated for various biodynamic activities.
Objective
To pursue a career in academics with research and development in the area of nano-sciences, as well as synthesis of novel bioactive molecules utilizing modern synthetic methods and collaborative medicinal chemistry studies.
