Christian Andrea Di Buduo has consolidated experience in research on megakaryopoiesis. Since 2009, his main research interest has been focused on unraveling how soluble factors, extracellular environment, and intracellular signalings integrate to promote megakaryocyte functions. Specifically, he demonstrated that autocrine signals, together with TPO, activate ion flows and biochemical pathways that finely regulate platelet formation and megakaryocyte sensing of the extracellular environment, and that any alteration of these mechanisms may lead to pathologic phenotypes (e.g., Inherited Thrombocytopenias, Myeloproliferative Neoplasms).
In an effort to try to better translate these laboratory results to clinic, he contributed to the development of the first silk-based bone marrow tissue model, that can faithfully mimic the bone marrow niche for ex vivo production of human platelets to be used in transfusion and regenerative medicine (Patent n. WO2016022834 A1) and testing of human-specific drugs (Patent n. WO2021113830A1). Of note, these systems were successfully used to mimic pathologic megakaryopoiesis by taking advantage of in vitro differentiated megakaryocytes from patients.
