I did my PhD at the University of Strasbourg, at the Institute for Cellular and Integrative Neuroscience (U3212, France). I studied different aspects of the circadian clock in the rodent retina, and primarily
1- its tissue localization using bioluminescence recording of cultured cell layers isolated by horizontal vibratome slicing,
2- its properties such as its period, and
3- cell-layer coupling using a pharmacological approach.
My first post-doc, in the same institute, aimed at characterizing neurochemical changes in the hypothalamus of Siberian hamsters gestated and lactated in different photoperiodic conditions, by
1- comparing gene expression profiles (RNA-seq) of purified tanycyte cells (laser capture microdissection) and by
2- verifying the causal effect of TSH (thyroid-stimulating hormone) on the expression of the deiodinases Dio2 and Dio3.
During my second post-doc, at the National Eye Institute (NIH, USA), I investigated another aspect of time: aging. The goal was to decipher epigenetic changes (ATAC-seq) that lead to gene expression changes (RNA-seq), using flow-sorted mouse rod photoreceptors as a model. I also studied changes in chromatin accessibility during post-natal development in murine rods, and the human retina regulome.
I then joined the Neuroendocrine, Endocrine and Germinal Differentiation and Communication Laboratory (U1239, Rouen, France) as a research engineer to work on a project aiming at improving spermatid yield and quality in organotypic cultures of cryopreserved pre-pupertal mouse testis tissues by using Multiome (10X Genomics) to evaluate
1- chromatin compaction in germ cells and
2- faulty cell programs in all cell types.
I was also in charge of managing and characterizing transgenic mouse lines.
I am now a research engineer in the Medical Genetics Laboratory (U1112, Strasbourg, France) were I study ciliopathies and Bardet-Biedl Syndrome in particular. I look into
1- finding and characterizing new pathological variants and
2- developing therapeutic solutions such as gene therapy.
