I started my scientific activity in the area of vertebrate endocrinology, particularly on the roles of prolactin and prolactin receptors and also on thyroid and sex hormones in vertebrate physiology, using a fish species (the seabream) as a model . These studies were carried out at the molecular biology level and included the cloning of the cDNAs encoding hormones, receptors and carrier proteins, their characterization and tissue distribution in adult animals and during their early stages of development, and their regulation by steroid hormones.
Later on, when I joined the health sciences research centre (CICS) of the University of Beira Interior, I established two main research lines based on the study of sex hormones in the regulation of genes involved in cancer and neurodegeneration. These studies focused on breast and prostate cancer and Alzheimer Disease.
In relation to breast and prostate cancer, we have identified novel genes involved in both diseases and are currently assessing the importance of their regulation by sex hormones in tumour development.
In relation to Alzheimer disease, we have been investigating the regulatory mechanisms of transthyretin (which prevents amyloid-beta deposition) and metallothionein in the mammalian brain.
More recently, the foccus of my research is being dedicated to the regulation of the choroid plexus protein synthesis in response to sex hormones, and to the analysis of its implications in neuroprotection and neuroregeneration within the scope of the project “Microarray based identification of estrogen and androgen responsive genes in rat choroid plexus” PTDC/SAU-NMC/114800/2009 ).
In addition I am also setting up another research line to look at the crosstalk between the choroid plexus and the cerebrospinal fluid in response to brain insults such as amyloid beta toxicity and brain focal ischemia using the rat as an animal model and to identify the sex associated factors which are protective against these insults underlying sex differences in AD and stroke incidence and severity among men and women.
The main purpose of these research lines are the identification of new molecular targets or agents for therapeutic and early diagnostic purposes.
