My main research interest is glial cell biology in health and disease, with particular expertise in astrocyte ATP-mediated calcium signalling and the regulation of oligodendrocyte differentiation, using the rodent optic nerve as a model CNS white matter that is relevant to human cortical white matter and neurodegenerative diseases, such as multiple sclerosis and Alzheimer’s disease (reviewed in ‘Structure and Function of Oligodendrocytes’, in Neuroglia, 2013). I have spoken at international conferences on glia (e.g. IBRO, Euroglia, FENS and the Gordon Myelin Conference), and written reviews and book chapters on the subject, as well as two books on glial cell biology (Verkhratsky & Butt, 2007, 2013). My main recent focus has been on Kir channels in glia and GSK3beta signalling in the controlling oligodendrocyte differentiation; their detailed description is beyond the scope of this biography, but the background that underpins my work has been comprehensively reviewed in Chapter 5.4 of ‘Glial Physiology and Pathophysiology’ (Verkhratsky & Butt, 2013). Through a PhD studentship from the Multiple Sclerosis Society and using a combination of genetic and pharmacological loss- and gain-of-function approaches, we discovered that GSK3beta and Wnt drive oligodendrogliogenesis and myelination in the postnatal mouse brain in vivo(Azim & Butt, 2011; Azim et al., 2012; Azim et al., 2014a,b). Now, we are identifying the key targets of Wnt signalling that drive OP proliferation and differentiation and are deregulated in ageing. In addition, we are developing new mouse lines to examine the functions of Kir channels and GSK3beta signalling in oligodendrocytes, using funds provided by the BBSRC and University of Portsmouth (UoP). These transgenic mice provide a unique and powerful resource that will help this important area of research.
