● Biochemical analysis of Phospholipase D in lipid homeostasis in Drosophila phototransduction
My research has focused on revealing fundamental mechanism underlying the role of Phospholipase D (PLD) in regulating lipid levels. We find that photoreceptors have a light-dependent PLD activity that is required to maintain phosphatidic acid levels during G-protein coupled receptor (GPCR) activation and signaling. PLD works in coordination with retromer function and Arf1 activity to regulate apical membrane size.
Junior Research Fellow - 2011 - 2013
● Standardization of mass spectrometry based Lipidomics to identify and quantify different glycerophospholipid molecules involved in Drosophila phototransduction.
The project involved optimization of highly efficient extraction of different glycerophospholipids from cells and intact tissues of Drosophila and total lipid phosphate assay for efficiency checking and data normalization. Later, my research was focused on development of direct infusion mass spectrometry using high resolution Orbitrap technology to identify and quantify individual molecules of glycerophospholipids. During this period, I have successfully characterized lipid regulation by different enzymes involved in Drosophila phototransduction. This method identifies lipids based on class specific chemical formula generated from their monoisotopic masses, so the exact composition of the fatty acyl chains could not be determined. To resolve this issue, I have developed a novel mass spectrometry-based method which couples LC-separation, multiple reaction monitoring and enhanced product ion scan to identify and quantify individual molecular species of glycerophospholipids with the exact composition of fatty acyl chains in sn-1 and sn-2 carbon (regioisomers).

Thesis Title: Preparation & Characterization of Mesoporous CeO2-ZrO2 Nanopowders using Dodecylamine & Sodium Dodecyl Sulfate as a Surfactant.
The work involved synthesis and analysis of the structure of Zr4+ doped mesoporous CeO2 nanopowders using different surfactants under different condition. The XRD data showed that mesoporous CeO2-ZrO2 nanopowders were sunthesized in cubic fluorite structure. FTIR results showed that the material was surfactant free and SEM analysis confirmed phase transformation.
