Many of the drugs currently taken are active at proteins that sit on the surface of cells called G protein coupled receptors (GPCRs). These drugs include beta-blockers to treat heart disease, antihistamines to treat hay fever and drugs that relieve the symptoms of asthma. My laboratory is interested in how to make drugs that act on GPCRs specifically in the brain for the treatment of neurodegenerative disease such as Alzheimer's disease. Over the last year my laboratory has made substantial progress in this area. We have found that by developing a new type of drug called an allosteric modulator that acts on a specific GPCR (the acetylcholine M1-muscarinic receptor) we are able to not only rescue the loss of memory in a mouse model of neurodegeneration but importantly we have shown that we can slow the progression of the disease. We believe that this novel finding opens the door to developing these drugs for use in people. As such we have started major collaborations with pharmaceutical companies including Eli Lilly and with centres of excellence in Australia and the USA to ask whether we can develop the findings into new medicines for Alzheimer's disease.
