I graduated in Biology by Faculdade de Ciências da Universidade do Porto (FCUP) in 2016.During my internship, I performed molecular biology techniques in the plant model Arabidopsis thaliana, with the aim of knocking out arabinogalactan proteins expressed during embryonic development, under the supervision of Prof. Isabel Amorim. Then, I obtained a Cell and Molecular Biology master’s degree at FCUP, in 2019. My master thesis, named “An evolutionary approach to understand how interactomes change when expansions of the polyglutamine (polyQ) tracts occur: implications for the human polyQ diseases”, was developed in the context of the Phenotypic Evolution group under the supervision of Cristina P. Vieira and Jorge Vieira. I have developed an in-silico protocol to infer the interaction recognition signatures (IRs), and using ATXN1 as model, I infered how these IRs are different in wild-type and the expanded form (responsible for the SCA1 disease), as reported in the BMC Medical Genomics manuscript (DOI: 10.1186/s12920-019-0594-4), where I am a co-author.
Towards the end of my Master thesis work(December 2019),I got a scholarship for three months, under the supervision of Cristina Vieira,to perform proteomic analyzes in SCA1 mutant flies,on which I developed Drosophila manipulation skills.
Furthermore, I also worked on development of the protein-protein interaction database EvoPPI3. Besides, unifying PPI data from the main databases, it includes literature-curated PPIs that are excluded from the main PPI databases, but are available in ProteomeCentral and as supplementary material of published articles, including those based on paralogues in model species mutant strains. This work was presented in a conference paper in Practical Applications of Computational Biology and Bioinformatics 2022 (PACBB2022) with the title “EvoPPI2 : A Web and Local Platform for the Comparison of Protein–Protein Interaction Data from Multiple Sources from the Same and Distinct Species” (DOI:10.1007/978-3-031-17024-9_10). It was also presented at an international conference in V CINBIO Meeting 2022 with the title “EvoPPI2 : an updated version of a powerful Protein-Protein Interaction database aggregator and analysis platform”. This work, together with the proteomics performed for SCA1 flies, lead to a manuscript on the Journal of Integrative Bioinformatics with the title ”On the identification of potential novel therapeutic targets for spinocerebellar ataxia type 1 (SCA1) neurodegenerative disease using EvoPPI3”(DOI:10.1515/jib-2022-0056).
In the last two years, I addressed the developed in-silico approach for SCA3. I identified putative ATXN3 IRs that are different in the two polyQ forms. Some of these results were presented at two conferences, namely “Drosophila as a SCA3 model : inferences on the Josephin domain role” presented at II International Meeting Of The Portuguese Society Of Genetics(2021) where I was the speaker, and “The role of the Josephin domain in the ATXN3 protein interactions” presented at IJUP2021 (Investigação Jovem da Universidade do Porto 2021). During this period, I also performed phylogenetic analyses using all animal genomes available at NCBI, to understand the evolution of the ATXN3 gene family. This work also found support for the functional redundancy of other Josephin-like proteins, which was first hypothesized due to the lack of a neurodegenerative phenotype in ATXN3 knockout mouse and nematode models. These results are reported in Frontiers in Molecular Neuroscience, where I am a co-first author (DOI:10.3389/fnmol.2023.1140719;Representative Document attached).
Currently, I am affiliated with the Phenotypic Evolution group at the Instituto de Investigação e Inovação em Saúde (i3S), where I aspire to further continue my research in the field of polyQ diseases. A PhD grant would be instrumental to continue my work in this area.
